ABSTRACT

Abstract We investigated whether coconut milk protein (CMP) contributes to the beneficial effects of coconut milk consumption on cardiovascular health markers previously found in middle-aged rats. CMP was isolated and precipitated from dried fresh coconut milk, then gavaged (1 g/kg) to middle-aged male rats for six weeks; control rats received distilled water. Compared to controls, CMP caused decreased body fat and lipid accumulation in liver cells and the platelet count. CMP did not affect basal blood pressure or heart rate in anesthetized rats. Vascular responsiveness to phenylephrine, DL-propargylglycine (PAG), acetylcholine or sodium nitroprusside was unaffected, but vasorelaxation to glyceryl trinitrate (GTN) increased. Effects of ODQ on vasorelaxation to GTN were similar in both groups. Expression of blood vessel eNOS, CSE and sGC was normal. The cyclic guanosine monophosphate (cGMP) level of CMP-treated rats was normal but addition of GTN increased cGMP and NO concentration more in CMP-treated rats than in controls, an effect unaltered by addition of diadzin. Taken together, CMP appears partially responsible for the improvement in cardiovascular health markers caused by coconut milk in middle-aged male rats

ABSTRACT

O contador automático hematológico ABX Micros ESX 60 (Horiba Medical 2012) é analisador hematológico veterinário multi-espécie que realiza 50 contagens por hora, libera 18 parâmetros sanguíneos, além de fazer representações gráficas (histogramas) para leucócitos, hemácias e plaquetas. O objetivo deste trabalho é avaliar o desempenho do referido aparelho em relação à linearidade e carryover, através de controle comercial e de amostras de sangue provenientes da rotina do Laboratório de Patologia Clínica Veterinária. De acordo com resultados é possível afirmar que o presente aparelho possui um excelente coeficiente de linearidade (r2=0,99) nos parâmetros de leucócitos, eritrócitos e plaquetas em relação às diluições estudadas. Em relação aos carryover houve excelente desempenho do aparelho, contudo, houve valores não conformes nos parâmetros de CHCM e VPM em uma das metodologias realizadas que pode ser justificada pela limitação da fórmula que não considera a características do equipamento.(AU)


The automated hematology counter ABX Micros 60 ESX (Horiba Medical 2012) is veterinary hematology analyzer multi-species that carries 50 counts per hour releases 18 blood parameters, in addition to graphical representations (histograms) for leukocytes, erythrocytes and platelets. The objective of this study is to evaluate the performance of the apparatus with respect to linearity and carryover through commercial control and blood samples from the routine of Veterinary Clinical Pathology Laboratory. According to results we can say that this device has excellent linearity coefficient (r2=0.99) in leukocyte parameters, erythrocytes and platelets during that time dilutions. Regarding the carryover was excellent device performance, however, was not in conformity values ​​in the parameters of MCHC and VPM in one of the methodologies made that can be justified by the limited formula that does not consider the equipment characteristics.(AU)

ABSTRACT

BACKGROUND: For the diagnosis of essential thrombocythemia (ET), no single clinical or laboratory finding of diagnostic value is available and a differential diagnosis of other myeloproliferative neoplasms or reactive thrombocytosis (RT) is needed. Following recent developments in automated blood cell analyzers, various platelet indices can now be measured. In this study, we analyzed whether platelet counts and 6 platelet indices can be used for the differentiation of ET from RT in patients with a platelet count of 600x10(3)/microliter or more. METHODS: The subjects studied were 31 patients with ET and 224 patients with RT. The platelet counts, mean platelet volume (MPV), plateletcrit (PCT), platelet distribution width (PDW), mean platelet mass (MPM), mean platelet component concentration (MPC) and large platelets (LPLT) were measured by ADVIA 120 (Bayer Diagnostics, USA). The mean values of each item were compared between the two patient groups and the sensitivity and specificity of each item in the diagnosis of ET were determined by ROC curve analysis. RESULTS: In essential thrombocythemia, all parameters except MPC were significantly higher than in reactive thrombocytosis. For the diagnosis of ET, the sensitivity and specificity were: 74.2% and 84.4%, when the platelet count was > or = 820x10(3)/microliter; 80.6% and 80.0%, when the plateletcrit was > or =0.63%; and 64.5% and 99.1%, respectively, when LPLT was > or = 23x10(3)/microliter. CONCLUSIONS: The platelet counts and platelet indices are useful for the differential diagnosis of thrombocytosis. The plateletcrit and LPLT are particularly useful for the diagnosis of ET when the platelet count is markedly increased.

ABSTRACT

Devido à tendência das plaquetas em aderirem nas superfícies estranhas, além da facilidade destas em aglutinarem e desintegrarem, contagem de plaquetas realizada por método manual apresenta certa dificuldade operacional, estando assim sujeita a resultados errôneos. Presente estudo teve como finalidade, verificar o efeito dos materiais mais comumente utilizados na colheita e diluição das amostras sangüíneas, sobre plaquetometria. Foram analisadas amostras de sangue de 18 indivíduos adultos, colhidas simultaneamente em recipientes de vidro e de plástico contendo EDTA 10%. Diluição destas amostras foram realizadas em tubos de plástico e vidro com oxalato de amônio 1% e a contagem realizada em câmara espelhada de Neubauer. Resultados apontadaram que, faixas plaquetárias obtidas de sangue submetido a diferentes tratamentos não apresentaram diferenças estatisticamente significativas, indicando por conseguinte que, tanto vidro quanto plástico podem ser utilizados sem restrições para realização deste exame.

ABSTRACT

The accuracy and precision of platelet counting using new automated blood cell analyzers is satisfactory. However, there are several errors in automated platelet counting. Careful review of blood cell histograms and peripheral blood films is necessary to avoid false platelet counts as well as to ensure a good quality control regimen.

ABSTRACT

La pseudotrombocitopenia es un fenómeno "in vitro" que se presenta diariamente en los laboratorios, como una de las interferencias en el cómputo de plaquetas, al realizar los hemogramas automatizados, de un pequeño porcentaje de la población. Su no reconocimiento puede llevar a consecuencias diversas en el manejo de los pacientes portadores, que no tienen sintomatología alguna por esa condición. En esta revisión se analiza cómo es que se produce el fenómeno, de origen inmunológico, cuáles son las implicaciones de su expresión y se dan pautas para identificarlo y proceder adecuadamente en los casos que se detectan en diversas condiciones.

ABSTRACT

Algunos autores han propuesto que una máquina de recuperación intraoperatoria de sangre, que posea la capacidad de modificar su velocidad de centrifugación a fin de ajustarla a la requisitoria de la separación de componentes sanguíneos, puede ser utilzada con este fin. La Stat-Compact de Dideco cumple con este requisito, por lo que analizamos sus posibilidades como procesador celular. Plasmaféresis terapéutica : dos pacientes con esclerosis en placa fueron tratados con 3 y 5 recambios plasmáticos respectivamente, obteniendo una disminución de la IgG en LCR (de 30,9 a 2,3 µg por ciento y de 38,9 µg por ciento a "no dosable") y evolución clínica muy favorable. Una paciente embarazada, portadora de un anticuerpo anti-D con un dosaje ponderal de 40,28 mg/ml, fue sometida a 6 recambios plasmáticos (con disminución de 28,5 mg/ml), interrumpiéndose el tratamiento por muerte fetal intraútero. En este aspecto, consideramos que, a pesar de obtener resultados auspiciosos (de acuerdo con la indicación), el número de casos no es suficiente para obtener conclusiones.

ABSTRACT

Thalassemias and hemoglobinopathies in Thailand have been examined with a blood cell counter based on electroimpedance principle and obtained size distribution curves of red cells and platelets. Among various disorders, beta-thalassemia/hemoglobin E and homozygous hemoglobin Constant Spring showed severe anemia. Their red cell size distribution curve shifted to the left and overlapped with the platelet size distribution curve. Red cell distribution width expressed by coefficient of variation and the degree of the overlapping were stronger in beta-thalassemia/HbE than HbH. Heterozygous beta-thalassemia showed a narrow red cell distribution curve width with small standard deviation and low England's value. Although the overlapping of size distribution curves cause inaccurate red cell count and platelet count, careful observation of the size distribution curves was proved to have high diagnostic value.
